In the process of plant breeding and production, the quality of pollen is related to the later seed yield and crop yield, and precise analysis of pollen characteristics plays an increasingly important role.At present, the most important method for studying pollen is flow cytometry analysis methods based on fluorescence staining or direct observation by microscopy. In response to these situations, Amphasys developed the Ampha Z30, a pollen mass analyzer that uses impedance flow cytometry analysis technology, opening a new chapter in pollen research.The instrument greatly shortens the sample pretreatment time, and the core microfluidic analysis chip that measures the fast analysis speed and does not require dyes, greatly saving scientific researchers' time and expenses.At present, the Netherlands Ansa Seed Company has widely used Ampha Z30 to study pollen characteristics.

Technical Principles
The core of the analysis of the pollen mass analyzer Ampha Z30 is a microchip based on the principle of Kurt counting.Compared with traditional Kurt counters, its advantages are small size and light weight (8kg).In the figure below A, after the cells (or granules) are bidirectional electrophoresis in the injection area (20x200um), the cells (or granules) are gathered in the center of the flowing liquid and arranged in a row; then the cells (or granules) pass through the red box.The marked measurement area (channel from 10x10um to 40x40um) has two different electric fields in this area. When the size, shape, and even the cell viability status of different cells (or particles) will be different, different electrical signal differences will be generated (as followsB and C in the figure, thereby calculating the number, activity and other parameters; the monitored cells (or particles) finally pass the sorting area, where there is an electric field in the opposite direction to the injection area, and the sorting area can be sorted. The cells (or particles) can be sorted.

At the heart of the Ampha Z30 cell counter is a microfluidic control chip equipped with microelectrodes that can be used hundreds of times.When cells or other particles in the solution pass through a specific electric field, the chip can analyze the characteristics of the particles by detecting the resistance flowing through the liquid.To ensure measurement sensitivity, Amphas has developed chips with different channel sizes from 15-250um, covering measurements ranging from small objects (such as bacteria) to large objects (such as pumpkin, corn and cotton seeds, etc.).
